Whether it is possible to do that depends on the controller.
Most USB controllers use protocols that are not supported by Audacity (such as MIDI or DMX). The only kind of USB controller than might work with Audacity is one that emulates keystrokes, and it would need to be configurable to match Audacity’s keyboard shortcuts.
