Receiving ”muffled” audio interference using Direct Monitoring during recording Mono audio using AUDACITY 3.7.7 and Focusrite Scarlett 2i2 interface with headphones for monitoring audio.

Direct monitoring doesn’t go through the computer.
If it’s a directional “studio” microphone, make sure it’s pointed in the right direction.

Yes, indeed, Audio Enhancements MUST BE TURNED OFF from ALL input devices (mics, Realtek, interfaces, etc) in order to eliminate “muffled” audio during Direct Monitoring, using headphones.
But, this ADVANCED function is buried DEEP INSIDE:
SETTINGS-> SYSTEM → SOUND → INPUT → DEVICES-> ADVANCED
