You can often get away with having a short amount of silence if the loop points (start and end of the file) are at zero crossing points http://en.wikipedia.org/wiki/Zero_crossing particularly if the start and end are at "silent" points in the loop. For example, if it is looped speech and the loop points occur between words, then a slightly extended gap between the last word and the (repeated) first word will be hardly noticeable.
