What's your setup?
XP / Vista?
Desktop / Laptop?
Sound card?
USB audio device?
Audacity 1.3.5?

Too much information is better than not enough.

If you are using a USB audio device, make sure that it is connected, switched on and recognised by Windows before you open Audacity.
If you are recording through your sound card, do not connect any USB audio devices to your computer.
Also, check the Audacity settings in "Edit > Preferences > Audio I/O" and "Edit > Preferences > Quality"

I use XP, a desktop, and a USB audio device called Logitech AK5370. It supports the project rate that I use, and it still doesn't work. I use Audacity 1.3 Beta (Unicode)

It started not working out of no where!

Make sure that the sound card is connected, switched on and recognised by Windows, then open Audacity.
Look in "Edit > Preferences > Audio I/O" and "Edit > Preferences > Quality" - What is selected for the Recording device? What is the default sample rate and bit depth set at? How many recording channels are set?

If necessary, correct any of the above settings, then close and restart Audacity so that the changes will take effect.
