Audacity will not work into a full or badly fragmented hard drive. You need to keep at least 20% free space to do live audio (or video) production.
Defrag
-- Right Click Start > Explore > Right Click Local Drive C: > Properties (Used, Remaining)
-- Tools > Error Checking & Defragmentation

Virus Protection, Email, Photoshop? There can't be any competition for the computer time, either.
You could have a ratty digital connection and that will create problems. Replug everything once or twice. Is it actually clear, missing musical notes, or is there noise, too.

If you open Audacity and go to "Edit menu > Preferences > Devices" there are 2 options for "Host". Try the other option.
