It is possible...

If your connection is mic -> mixer -> line-in you have to select line-in as the input to audacity.

1) select line-in as the recording device in audacity
2) import the mp3 track to audacity
3) set overdub ON and software/hardware playthrough OFF
4) turn off your speakers and listen with headphones
5) hit the record button and sing to the mic

You should be listening to the mp3 on your headphones and a new track should be recording your voice online.
Include as much details as you can in your post (Audacity version, Operating System, Equipment used, etc).
