I often work with large single tracks (10 hours). Manipulation of these (eg amplification) often causes Insufficient Disk Space errors. Is it possible to turn off the undo & would this resolve the issue?

No it is not possible to turn off the Undo history.
You could save and close the project after each operation (closing the project deletes the Undo history), or better would be to split the huge project into more reasonably sized projects and work on each part separately and/or batch process with macros.
