While you can probably force that to work, it complicates things immediately if Something Goes Wrong. Audacity likes being installed with all the other Windows programs on the system drive.

Do you use Skype or other chat, communications or sound program? Skype in particular likes to change settings to suit itself when it feels like it, and it’s not unusual for Audacity to get left out in the cold.
One note, Audacity checks for devices and microphones when it starts. If you start and then plug in, Audacity will not be able to find your microphone. Restart Audacity or Transport > Rescan.
I’ve been ignoring the loud, clanging alarm bells until now, but if you installed Audacity on a thumb drive because you have no room on the system drive, your chance of a long, happy editing career is zero. Audacity does a lot of production by managing sound in real time and because of the way it manages UNDO, shows get really big in a hurry. Running from a USB drive on a system starved for resources is not recommended.
