Sure! If you are recording in stereo, you should be able to get the guitar on one side and vocals on the other side. Then, you can split those into separate files if you wish. The Amplify effect should work just fine for what you're trying to do... If you have a LOT of patience and a good ear, you might be able to replace the note(s) with the same note from a different part of the performance. You'll need to start/end your selction on a waveform zero-crossing, or you may need a short (millisecond) crossfade in order to get a smooth transition....
