In the Edit menu, select "Preferences" and on the "Audio I/O" tab switch off "Software playthrough".

Try setting the default Quality settings to 16 bit, 44100 Hz and see if you still have the problem (Edit > Preferences > Quality).
You can also try zooming out to show the entire track so that Audacity does not need to scroll while it plays.

Try using Audacity 1.3.5 and make sure you start a new project each time you want to edit a file.

How are you playing the karaoke - on your computer or from an external karaoke player?

Are you able to pan the karaoke music to left or right? Are you able to pan your vocal to the other side?
