This is a limitation of the MP3 format - there is always a small amount of "padding" at the beginning of an MP3 file.
To avoid this problem, use a different format such as WAV, FLAC or OGG.

On Mac I'd suggest trying the AU version of AnSpec rather than the VST.

There is no mention in the Quick Start Guide of the mixer having USB audio, so I'd assume that it does not have USB audio.
