If the vocal and only the vocal is dead centre in the mix , then the "Blue Cat Widening Gain" which is available as part of the free "Gain Suite" http://www.bluecataudio.com/Products/Product_GainSuite/ can isolate the vocal. If that is not the case then it probably can't be done...

Most of the effects have parameters that reset on restarting Audacity.
"Preferences" are intended to persist so that you don't need to set them to the way that you prefer each time you use Audacity (as is the case with almost all other software).
Which settings in particular do you mean?

On some machines you can combine two USB devices in the "Aggregate" service, That functionality is not built into Windows, only Mac and Linux. i tried to change in the audio i/o but the audacity say that the play card and the record card should be the same. What was the exact error messag...

The manual is not included in the ZIP version of Audacity 1.3.x (this is to make the ZIP version a smaller download). The manual is included in the .EXE installer which is available here: http://audacityteam.org/download/beta_windows

Audacity 1.2.6 is not suitable for Windows 7 as it will often crash when you stop the recording. The correct version for Windows 7 is 1.3.13 (this is due to be updated to 1.3.14 in the next few days). You can create multi-channel files with Audacity.
The file can then be played in any media player that supports the file type that you create.

AMR files should be supported by current versions of Audacity 1.3 provided that you also have FFMpeg installed and Audacity is correctly configured to use FFMpeg.
