Which version of Windows?
Check the settings in the Windows Control Panel to ensure that the recording level is turned up there and it is not muted.

Audacity does not currently support the BWF format, so track marks are not supported.
Audacity does have "label tracks" which allow "markers" to be used within an Audacity Project. http://manual.audacityteam.org/man/Label_Tracks

Check that you have a good input level on the Tascam.
Turn up the level until you just get an indication on the clip indicator, then back it off a short way.

To apply an effect, Audacity needs to be stopped (not in Playback, Record or Pause).
Ensure that the track that you wish to apply the effect to is selected.
To use the Noise Removal effect, the effect must have a Noise Profile before the effect can be applied.
