I presume that you tried rebooting the computer?
Check in the Windows Sound Control Panel that the sound card has been detected by Windows, is enabled and working - If Windows does not see the audio device then Audacity will not be able to.

First, I notice that the (s-rest 1) expression does not return exactly zeros. Nyquist, like other effects in Audacity, processes in 32 bit float. It is highly recommended that you work in 32 bit float format in Audacity (unless you have a really good overriding reason not to ;)) 32 bit float format...
