I would generally recommend using "MME" rather than "direct sound" because "direct sound" is only emulated in Windows Vista and later.
(It probably won't make any difference, but just as a general point ;) )

Your headphones need to be plugged into the U-Phoria, and select the U-Phoria as the playback device in the Device Toolbar.

What sort of microphone are you using, and how is it connected to your computer?

With a normal computer mic, it is possible to monitor the mic input, but there's a delay which is very distracting and makes it nearly impossible to concentrate on what you are doing.
