Assuming that you are using Windows Vista or later, try using the "WASAPI loopback" method described on the page that Koz linked to (that method tends to be the most reliable, but is not available on Windows XP).

Try Exporting as a WAV file ("File menu > Export Audio").
Avoid using dots or other special characters in the file name.

As you have recently updated from 2.0.6 to 2.1.0, please recheck that it actually is 2.1.0 that is running by looking in "Help > About Audacity". Also, check (or re-check) that AU type effects are enabled in the Effect preferences (Edit > Preferences > Effects) http://manual.audacityteam.o...

Yes that is often the case. There are different schools of thought about this, but in my experience a little bit of processing can often make a big improvement, but a lot of processing can often do more harm than good.
