The first thing I'd suggest is to reboot the computer.
Ensure that your headphones are connected before you launch Audacity.

In the "Vocal Reduction and Isolation" effect, the "Remove Centre" option may be best as it is less reliant on phase correlation than the simple "invert and add" method.
