I've not tested, but I would not expect Melodyn to work with Audacity. It is a very complex plug-in requiring comprehensive VST support, whereas Audacity's VST support is quite basic. Downloading the EXE version is usually the best option as this will install LAME in a location where Audacity can find it automatically.
