I don't see a Nyquist prompt available under Effect (it's available from Tools In Audacity 2.3.1, the Nyquist Prompt has moved to the "Tools" menu (it was previously in the "Effect" menu). but I got a touchy dialogue warning when I pasted it there and hit OK What's the warning?

I changed the semitones up and down, +10 and -10. 10 semitones is getting close to an octave (12 semitones in an octave). If you sang a C# instead of a C, then that's 1 semitone. If you sang a C a bit sharp (but not as sharp as C#), then you need to correct by a fraction of a semitone. Large change...

Assuming that the track is 32-bit float format (the default), then in normal circumstances no, there are no losses caused by the Amplify effect. Last time I looked, iTunes play lists could be exported as M3U files.
