You've probably still got the first project in play, record, or pause mode. "Stop" the playback in the first project and play should be available in the other.

The microphone input of your computer is probably mono. If the microphone input is mono, and your cassette tape is stereo, then the microphone input will only hear the right channel. Ideally you should use a cassette player that has "line level" stereo outputs, and an audio device / sound ...

Perhaps you have an anti-virus program checking the audio data before it allows Audacity to access it.
