The headphone uses the connection in the top illustration and the microphone uses the connection on the bottom.

Audacity saves Projects which are multiple production files that make up your show. To get an actual sound file, you need to Export one. File > Export As WAV.

You can have a very extended Audacity startup if you have a very large collection of plugins. One or two badly coded plugins can bring Audacity to its knees.
Edit > Preferences > Effects > Deselect all the effects and restart Audacity. Let us know.

Is Audacity running minimized? Is there an icon on the "Start" menu bar along the bottom (assuming you didn't change it)? It's not supposed to wake up minimized, but I guess anything possible.

To answer your specific question, yes you can daisy chain microphone amplifiers, but don't bet the farm on good results. the second one will amplify all the noise of the first one.
