Did you just recently use it on Skype, or have you been using it on Skype for years? Skype installs tend to screw up microphones and sound management to suit themselves. Make sure the Windows Control Panels are still pointed to the right microphone/devices, and then go into the Audacity Preferences ...

No. You can't take a finished show apart into individual voices or instruments. You can't change the volume of one and not the others. Sometimes in very high quality (music CD) stereo shows, you can remove the singer with Effect > Vocal Removal. But most of the time that doesn't work, either. Koz
